右美托咪定抑制HMGB1/TLR4/NF-κB信号通路减轻大鼠心肌缺血再灌注损伤研究

【摘要】 目的:观察右美托咪定(Dex)对心肌缺血再灌注损伤大鼠心肌功能、炎症反应及HMGB1/TLR4/NF-κB信号通路的影响。方法:取75只成年SPF级10周龄雄性SD大鼠。运用可复性结扎LAD法建立心肌缺血再灌注损伤(MIRI)模型。随机均分为假手术组(Sham组)、Dex预处理+MIRI组(Dex组)、Dex+育亨宾+MIRI组(Dex/Yoh组)、育亨宾预处理+MIRI组(Yoh组)与MIRI组(I/R组),每组15只。比较各组心肌梗死面积(IS/AAR)。运用ELISA检测并比较各组心肌组织与血清TNF-α、IL-6水平。采用 Western blot检测并比较心肌组织HMGB1、TLR-4、MyD88、NF-κB蛋白表达情况。结果:Dex组、Dex/Yoh组、Yoh组及I/R组心肌组织与血清TNF-α、IL-6水平均高于Sham组,其中Dex组
