针刺对脑缺血损伤大鼠海马区Bck-2、Bax蛋白表达的影响

     摘要目的:探讨针刺对高血脂合并脑缺血损伤大鼠海马区Bcl-2、Bax蛋白表达的影响。方法:雄性SD大鼠,随机分为空白对照组、脑缺血模型组、针刺治疗组,每组10只。采用化学刺激诱导血栓性闭塞大脑中动脉造成大鼠局灶性脑缺血模型。“针刺百会”、“水沟”,每日1次,治疗7d,治疗结束后,应用免疫组化方法观察针刺前后大鼠海马区Bcl-2、Bax蛋白表达的变化。结果:大鼠局灶性脑缺血后Bcl-2表达降低,Bax表达升高,Bcl-2/Bax比值下降(均P<0.01);针刺后脑缺血大鼠Bcl-2表达增加(P<0.05),Bax表达降低(P<0.01),Bcl-2/Bax比值升高(P<0.01)。结论:针刺能增强Bcl-2的表达,抑制Bax的表达,这可能是针刺减轻脑缺血损伤的机制之一。

    Abstract Objective: To observe the effect of acupuncture on Protein Bcl-2 and Bax expression in hippocampus in cerebral ischemia (CI) rats. Methods: Thirty male SD rats were randomized into control, CI, and CI + acupuncture groups, with 10 cases in each group. CI model was established by occlusion of the unilateral middle cerebral artery. “Baihui”(GV20) and “Shuigou”(GV26) were punctured and stimulated by twirled the acupuncture needle with hand continuously for 1 min and to maintain the needle for 20 min in point “Baihui”. The treatment was conducted once daily and continuously for 7 days. The expression of protein Bcl-2 and Bax in the hippocampus was displayed by using immunohistochemical method. Results: In comparison with normal control group, Bcl-2 immune reaction(IR) positive cells in the hippocampus in CI group decreased significantly(P<0.01).Bax IR positive cells in the same group increased significantly(P<0.01),while Bcl-2/Bax also reduced remarkably(P<0.01),meaning that after ischemia the expression of Bcl-2 was downregulated and Bax upregulated markedly. In comparison with CI+group,Bcl-2 IR activity increased, Bax IR activity reduced and Bcl-2/Bax increased considerably in CI+ acupuncture group(P<0.01,0.05), suggesting that acupuncture could reverse the effects of CI. Conclusion: Acupuncture can eliminate cerebral ischemiainduced abnormal expression of Bcl-2 and Bax in rats with cerebral ischemia. ......
