靶向核转录因子-κB的siRNA和甲氨蝶呤纳米脂质体对类风湿关节炎大鼠骨破坏的影响

【摘 要】目的:探讨靶向核转录因子-κB(NF-κB)的siRNA和甲氨蝶呤(MTX)复合纳米脂质体对胶原诱导类风湿关节炎模型大鼠骨破坏的影响。方法:将48只健康SD大鼠随机分为空白对照组10只和造模组38只。造模成功的30只大鼠随机分为模型对照组、MTX组、MTX复合纳米脂质体组
(si-MTX组),每组10只。测量各组大鼠治疗前后体质量、脏器指数、足趾肿胀度、血清中炎性因子水平,以及滑膜组织中相关基因和蛋白的表达水平。结果:MTX和复合纳米脂质体均可显著降低胶原诱导性关节炎大鼠胸腺指数、脾指数、足跖肿胀度,降低血清中炎症因子水平,调节滑膜组织中相关基因和蛋白的表达(P < 0.05),且si-MTX组作用更强(P < 0.05)。结论:复合纳米脂质体发挥作用的机制可能是阻断了NF-κB通路,减少促炎因子释放,抑制骨吸收,保护关节软骨,且靶向性、安全性更高。
